How do you conduct a background check on someone especially a guy and his family before approving their rishta for your daughter/sister,etc? What are the ways of conducting background check?

And then there is computer surveillance, aerial surveillance, human intelligence (HUMINT operatives), satellite/GPS tracking and so forth.

No this is really important in the rishta process! Often people will come and put on q big show pretending to be something they're not, but once you get to know them you realise their lies! It can make or break a rishta!
I'd say find out from people who know him well but are not close enough to lie for him, so like people who maybe know him in a professional capacity e.g. work colleagues, as well as family members and his friends.

most families can be traced back to Pakistan/India where SOMEONE tends to know them. And stuff usually comes out if they're lying. From my experience and from seeing my friends/cousins get married, most "shady" people will run for their lives as soon as you ask for "references". That's a red flag...So even just bringing that up should deter most liars/deceivers.

My best friend's family got some rishta from Pak. THey kept saying that the guy had studied here so my friend's family was alright, we'll consider them. They kept asking for friend's pictures and things got to the point where the honay wali saas was asking her about what color her engagement/wedding outfit should be, all the while ignoring friend's parents' request to 1) show them pictures of the guy at least 2) let the guy talk to the girl 3) do investigation. In the girl's family's mind, things hadn't gotten to the "let's talk specifics about wedding yet" but the boy's side KEPT pushing. Finally my friend's dad put his foot down and said that they weren't gonna budge because it was slightly ridiculous that they hadn't even seen the guy's picture at this point so the larkay walay needed to slow their roll. Well the guy's side vanished and we heard through a third party that it was because all of a sudden the istikharah came out negative. My family knows people in Pak that are from the same city as the boy so we decidd to investigate. Turns out guy was morbidly obese, 5 years older than what they had told my friend and his "schooling in America" consisted of community college classes on the east coast...needless to say, she dodged a bullet.
it's always sketch when the family (ether boy or girls side) seems to rush into making a commitment. Most people will WANT to do research before handing over their son/daughter (especially true in arranged marriage settings where there's SO MUCH emphasis on family). And major red flag seems to be when they try to do the whole "oh we don't care about stuff like that, that's so backwards and jahil". Chances are they've done their research already. Make sure you still do yours.

There was a proposal for my sister.The guy said he was a doctor,graduated from KEMU (Lahore).The family seemed nice and everything else looked good.My parents inquired about them.Got good reviews,that they are great people and great family and all that.One of my parents' friends had some family members who were neighbors with this family.Even they said that they knew the guy since he was a kid.My father asked another one of his acquaintances just randomly,and it came up that the guy never went to KEMU.He was a doc but went to Russia for med school.When they were questioned,they actually objected and said that they were 'insulted' that so many inquiries were being done about them bla bla.Needless to say,the rishta got rejected.

Blood test! Just wait until after the first rishta-meet and prior to your first phone-date before dragging him to the clinic.

Don't forget to stick a GPRS tracker on him as well to make sure you can check that he doesn't go to any naughty places

Sorry, but that just isn't workable enough. Unless you're going to be marrying a nerd that has no idea of how electronics work. The rest could simply attach the GPRS module to a external 12V+ source located near a library that Einstein used to dig through prior becoming a profound genius and call it a day.
