They were NOT trying to find him. They had found him already through satellites that pick up communication, (ISI had provided a number and they knew that number belonged to someone close to OBL), they had satellites and drones that took real time images and followed the courier to the house, but they still did not know it was him as he hardly left his room, except to take a walk in the garden after dark. Therefore they wanted to get the DNA to confirm as they were unsure if it was really him.

They assumed they had found his location but they needed proof that he was actually there and thus they recruited Afridi to run a vaccine campaign to collect DNA.
