one of my friend's kid brothers used to play with her barbie dolls all the time! he was more attached to them than she was but so what, it didn't have a negative effect on him at all. i don't think its a sign of anything other than him transferring his affections on something else, perhaps mimicking you, and all the other reasons stated here. and keep in mind, he is the middle one- perhaps he just wants something of his own, all his own, that he can lavish attention on and have it be something exclusive to him. i'll bet with three boys theres probably a lot of competition for toys and space and attention from you and your hubs going on as it is-- maybe this is "his thing" that only he plays with and is responsible for.
